one doubt, STR are called microsatelites so arntt ehy supposed to be in the secomdary constrictions , how can they be found in the loci? this is a wonderful video BTW i love these animnations and explanations, love from india

i'm still confused what the point of STRs or VNTRs is in the process of DNA fingerprinting. like, i understand how use of restriction enzymes is relevant: specific restriction enzymes work by cutting at a specific base sequence, and because everyone's genome is unique, different people will have these base sequences in different spots, leading to the enzyme cutting at different spots, leading to different lengths of dna in different people. these different lengths in different people would show in electrophoresis as different banding patterns which show the two sets of dna compared are from different people. unfortunately people in the comments seem to be saying the use of restriction enzymes is an old technique and using strs and PCR is the ore efficient way to go. unfortunately i cant see how you can tell the difference between two people by amplifying their strs and separating them using electrophoresis? like ok lets see, we have two people we wanna compare, we get their dna somehow and we find the loci of their dna that varies the most between different people. im gonna take a guess and say that the reason it varies the most amongst different people is because different people have different numbers of strs in this loci? well ok, but then wouldnt it be likely that two different people have, lets say, 13 strs coincidently? and even if it werent likely, and they did have different numbers of strs, where would i go next in this process? do i amplify this segemnt that has the 13 or whatever number of strs using pcr and then run the segments through electrophoresis? i guess if two people were different and had a different number of strs at this loci then one band would show up on each persons gel in different locations down the gel, but im pretty sure the result is multiple bands as you tend to see on majority of the dna fingerprints.... so does that mean that this loci we've been looking at has multiple sets of strs that we somehow segregate into separate strands, amplify them in pcr and then run them through eectrophoresis? but wouldnt that mean we need to use some restriction enzyme? welp.
