What are the limitations of the challenge in selecting perks? Such as, seems like pretty stock start, not a lot of negatives to grab positive perks

How many deaths allowed, or always starting a new complete one?

Really bad luck with infection you've got here. I've been doing this challenge in 42.5 for 14 in game days, cleared to 12th floor, almost 1000 kills, god knows how many dropped from the edge. Been lacerated and scratched 5-6 times, lucky so far. But I think the worst is yet to come on the bottom floors...

I would look into the proximity inventory mod for this challenge. You are going to have giant piles of corpses that you will want to loot, and looking through each corpse's inventory is going to be a nightmare

Targeting in build 42 is definitely different. Even with the targeting outline on, sometimes it just misses the target. Might be good to just turn the outline back on and re-learn.
